To understanding the miRNA expression profiling of cancer stem cells of laryngeal squamous carcinoma, the total RNA of CD133+CD44+ laryngeal cancer stem cells (isolated from LSCC cell line TU-177, named TDP), CD133-CD44- cells (TDN) and parental TU-177 (unsorted TU-177 cells, named TPT) was extracted, followed by miRNA sequencing. Differentially expressed miRNAs were identified. Overall design: The CD133+CD44+ cells (TDP) and CD133-CD44- cells (TDN) were isolated from TU-177 cells and cultured. Total RNA was extracted using TRizol reagent (Invitrogen, Carlsbad, CA) following the manufacturer’s instruction. Equal quantities of RNA from 3 samples in one group were mixed. The small RNA library was prepared. The library products were finally sequenced on Illumina HiSeq 2000 platform.
