This dataset contains mechanical measurement of tamoxifen (TAM)-inducible MCF10A-ER-Src cell line (Hirsch et al., 2009, Cancer res.) conducted using atomic force microscopy (AFM).
Triplicates of the following experiments are included:
• EtOH vs TAM - tamoxifen induction over 72 h vs uninduced control (EtOH)
• EtOH-CAV1KD - CAV1 knock-down in EtOH-treated, uninduced cells
• TAM-CAV1OE - transient overexpression of CAV1 in TAM-induced cells
Measurements were performed using atomic force microscopy (AFM) with a followings setup: Nanowizard 4 (JPK Instruments, Germany).
Tip-less cantilevers (Arrow TL1, nominal spring constant k = 0.35–0.45 N m−1, Nanoworld, Switzerland) with a 5-µm bead (PS-R-5.0, microParticles, Germany) glued at the end were used as the indenter.
Indentations were performed with a piezo extension speed of 5 μm s−1 to a maximum set force of 2 nN.
Following sub-folders are included under each sample/experiment:
• calibration - contains data acquired during cantilever calibration (whenever available)
• *sample name* - indentation curves in .jpk-force format
• *sample name*_processed-yyyy.mm.dd-hh.mm.ss - results of processing of indentation curves
