This dataset contains raw single-molecule STORM (stochastic optical reconstruction microscopy) image stacks of Alexa Fluor 647-labeled microtubules in COS-7 cells, collected to compare the sealing performance of different materials. The data correspond to Fig. 3 of the manuscript "UV-curable resin as a rapid and superior sealant for STORM" (submitted to Biomedical Optics Express). The dataset includes TIFF stacks from four sealants (UV resin, 5-minute epoxy, CytoSeal 60, and nail polish) at three time points after sealing (1 h, 12 h, and 48 h). Each file is a multi-page TIFF containing 40,000 frames at 50 × 50 pixels. These cropped regions of interest retain full temporal information while reducing file size for sharing. Acquisition parameters: - Pixel sampling: 130 nm per camera pixel - Camera: Andor iXon Ultra 888 EMCCD (EM gain 50) - Exposure time: 20 ms per frame - Camera conversion factor: 5.16 electrons/ADU - Camera baseline offset: 510 ADU - Image depth: 16-bit These raw data are intended for transparency and reproducibility, enabling independent validation of single-molecule blinking behavior and quantitative image reconstruction under different sealing conditions.
