ONT direct RNA sequencing of MCF7 cells upon E2 induction

After hormone deprivation (HD) for 72 hours, MCF7 cells were treated with 100 nM 17 b-Estradiol (E2) for 45 minutes, 3 hours, and 12 hours. Total RNA was isolated with TRIzol reagent and poly(A) enrichment was done (Dynabeads Oligo(dT) #61002). MinION Mk1C and direct RNA sequencing protocols were used as suggested by Oxford Nanopore Technologies (ONT) with Direct RNA Sequencing Kit (SQK-RNA002) and Flow Cell (FLO-MIN106D) to investigate the effect of E2 induction on the MCF7 breast cancer cell line transcriptome.
