HT1080 cells were grown on collagen I and treated with PI for 4 h (short term) and 24 h (long term) while RNA was extracted for Genome-wide Affymetrix Microarray gene expression analysis. The objective of this study was to identify whether collagen accumulation driven by blocking proteases induced transcriptional changes that correlated sustained amoeboid cell state. Overall design: 9 samples (3 x 3 biological replicates). Cells were plated on deformable matrices. Human fibrosarcoma cells (HT1080 cells) plated on deformable matrices were left untreated or treated with proteinase kinase inhibitor cocktail.
