SCALE2019Spring_Sequencing

Seawater samples were collected from both northbound and southbound transects in the Atlantic sector of the Southern Ocean on the SCALE 2019 Spring cruise. The transect went from Cape Town, South Africa to the ice edge near the South African Antarctic base and back to Cape Town. Only surface samples were collected on the southbound transect and both surface and chlorophyll maximum samples were collected on the northbound transect. Two incubations with waters from the mixed layer were also performed. Samples were collected onto 0.2 micron sterivex filters. 16S rDNA and 18S rDNA amplicons were sequenced from DNA extracted from all samples. RNA was also collected and sequenced from select samples.
