Effects of SOD and L-NMMA against DJ-1 oxidation induced by 6-OHDA. (A) Cell samples treated with 600 µM 6-OHDA in the presence or absence of 10 µg/ml SOD for 3 h were separated by 2D-PAGE and subsequently subjected to western blot analysis using the anti-DJ-1 antibody. The ratio of oxidized DJ-1 is shown under each condition (n = 2). (B) Cells were pretreated with or without 500 µM L-NMMA for 3 h, and this was followed by treatment with 600 µM 6-OHDA for an additional 3 h; and the cell samples were separated by 2D-PAGE and subsequently subjected to western blot analysis using the anti-DJ-1 antibody.
