Circadian rhythms in NAD+ bioavailabity are present in many tissues. Also, a pronounced depletion in NAD+ levels is observed in diet-induced obesity. This study was designed to investigate transcriptional responses triggered by a timed treatment with NAD+ which recovers heptaic NAD+ oscilations in obese mice, and ameliorates their metabolic dysfunction. Mice were fed either a normal diet (CD group) or a high fat diet (53% Kcal from fat) for eight weeks. Then, high fat-fed mice were divided in two groups, one was intraperitoneally (IP) injected with 50 mg/kg NAD+ at ZT11 (HFN group), and the other was injected with saline solution at ZT 11 (HF group), every day for three weeks under the same feeding conditions. Livers for transcriptomic analyses were harvested either at ZT6 or at ZT 18 (n=3 animals per time point and group: CD, HF and HFN).
