Spermatogonial stem cells (SSCs) provide foundation for robust and continual spermatogenesis in mammals. We report here that transcription factor E4F1 is essential for sustaining a foundational SSC pool by regulating metabolism. Conditional deletion of E4f1 in mouse germ cells resulted in progressive loss of the entire undifferentiated spermatogonia.
