Among the key molecular players in colorectal cancer (CRC) progression is the Migration and Invasion Enhancer 1 (MIEN1) protein, which has been implicated in tumor invasion and migration in various cancer types. Despite its evident role in promoting cancer aggressiveness, the precise mechanisms by which MIEN1 drives metastasis in CRC remain poorly understood. Addressing this knowledge gap is critical for leveraging MIEN1 as a potential biomarker or therapeutic target. In this study, we sought to investigate the functional role of MIEN1 in CRC using a CRISPR-Cas9 knockout model in the HT29 CRC cell line. The knockout model was generated by deleting minimal promoter region, anticipating a reduced migration invasion potential in the knockout cells.
