Silencing of Ankrd22 altered the distribution of macrophage subtypes in the tumor microenvironment and attenuated their pro-inflammatory activity Ankrd22 knockout mice were commissioned for construction by Cyagen Biosciences using CRISPR/Cas9 technology. The F4/80+ cell subset, isolated from mouse peritoneal cells through FACS sorting, corresponds to the population of mouse peritoneal macrophages. Single-cell suspension was added to the 10x Chromium chip according to the instructions for the 10x Genomics Single Cell 3 v3 Reagent Kit, with the expectation of capturing 8000 cells. cDNA amplification and library construction were performed according to standard protocols. Libraries were sequenced by LC-BioTechnology on an Illumina NovaSeq 6000 sequencing system at a minimum depth of 20,000 reads per cell.
