We performed 3'single-cell RNA sequencing (scRNA-seq) using the 10x Genomics platform on Arabidopsis thaliana leaf protoplasts derived from SETI (Super ETI) transgenic lines. These lines enable inducible activation of effector-triggered immunity (ETI) via the expression of the bacterial effector AvrRps4. To investigate early transcriptional responses during ETI, we treated plants with estradiol (E2) and collected protoplasts at 4 hours post-infiltration (hpi). Mock-treated samples served as controls.
