Gene expression analysisi by next generation sequencer revealed osteoclast diferentiation after the addition of PC3M-derived extracellular vesicles (EVs) with or without RANKL. Finally, we found that PC3M-derived EVs promote osteoclast differentiation in the presence of RNAKL. Overall design: To examine the effect of PC3M derived extracellular vesicles on osteoclast differentiation, we performed RNA seq. After an initial attachment period for 12 hours, the RAW264.7 were cultured in the presence or absence of recombinant mouse RANKL (GST-RANKL, Oriental Yeast Co., Tokyo, Japan) at a concentration of 10 ng/mL alone or in combination with PC3M derived EVs (30µg/mL) for 48h or 72h. Total 16 samples were examined: only addition of PBS (48h and 72h), PC3M derived EV (48h and 72h), only RANKL (48h and 72h), and RANKL with PC3M derived EV (48h and 72h).
