The underlying image data and analysis scripts for Figure 3B-E and Figure S1 of Believing is seeing – the deceptive influence of bias in quantitative microscopy.
The file 20230519_Ptk2_live_TransferrinER_63X_cell3_Z52_airyprocessed.czi contains an image of fixed cells labeled for endosomes and endoplasmic reticulum (ER) as described in README.txt and in the associated publication. The relevant channels of this file are also provided as separate tif files (20230519_Ptk2_live_TransferrinER_63X_cell3_Z52_airyprocessed_ER.tif and 20230519_Ptk2_live_TransferrinER_63X_cell3_Z52_airyprocessed_Transferrin.tif) to simplify use of the associated MATLAB script (segment_measure_ER_endosomes.m). The script requires three subfunctions which are included (bpass3D.m, cntrd3D.m, and pkfnd3D.m). Expected output from the MATLAB script is also provided (analysisOutput.zip).
