This project compares the expression profiles of induced pluripotent stem cells either unedited or edited to carry a homozygous or heterozygous missense variant in the SART3 gene, found in four children from two families. The three cell lines are PCS.201.010 unedited control, PCS.201.010 edited to carry the heterozygous SART3.p.Arg836Gln variant and PCS.201.010 edited to carry the homozygous p.SART3.p.Arg836Gln variant.The aim was to understand how this variant might affect cell signalling. These were grown from frozen stock three independent times to create triplicate cell pools for RNA sequencing. RNA was extracted using the ReliaPrep RNA miniprep system (Promega).
