Transcriptome analysis of drought resistance of wild rice SSSLs during germination under PEG stress

To explore the drought resistance mechanism of wild rice SSSLs in germination stage. In this study, B32 was selected as experimental material in the peG-6000 simulated drought stress germination test with Hua jing indica 74 as receptor and Zhanyin wild rice as donor, and the roots and buds of B32 and HJX74 were analyzed by RNA-SEQ transcriptomic sequencing. Combined with bioinformatics analysis, 16,411 genes with expression differences of more than 2 times were screened, including 10,579 genes with up-regulated expression and 5,832 genes with down-regulated expression. Among these differentially expressed genes, 74 genes were differentially expressed in B32 and HJX74 roots, 169 genes were differentially expressed in B32 and HJX74 shoots, and 46 genes were differentially expressed in B32 and HJX74 lines and shoots at different treatment time points. Among them, 25 genes were annotated in GO functional database, and two genes were annotated in KEGG database. The intersection of differential genes induced by water stress in photosynthesis suggests that water stress may affect the photosynthetic process of crops, and glutathione metabolism pathway plays an important role in rice response to water stress. This result confirmed and supplemented the previous research results, and also provided a good foundation for the excavation and functional research of drought resistance genes in rice and other crops.
