The aim of the study was to obtain plasmids containing the SAT1 flipper knockout cassette together with the MET15 gene encoding O-acetylhomoserine sulfhydrylase, as well as the downstream fragment of the MET15 gene. The construction of such a plasmid enables complementation of the MET15 gene in the C. albicans deletion strain using the SAT1-flipper method.
Complementation of the MET15 gene enables the assessment of the impact of this enzyme on the cell phenotype and strain virulence, and allows the determination of the in vivo function of the tested protein in L-methionine biosynthesis.
