To investigate the role of SRSF2 in skin we made use of mice where SRSF2 can be conditionally deleted or a point mutation can be introduced in an undifferentiated population marked by expression of Keratin 14. Overall design: Mouse back skin was treated topically with 4-hydroxy tamoxifen for up to 28 days. Upon collection mouse back skin was snap frozen. In order to assay the transcriptomic changes caused by SRSF2 loss or mutant SRSF2 total RNA was extracted uing Trizol reagent, according to manufacturer's instructions. RNA-sequencing libraries were then prepared. Comparative gene expression profiling analysis was then performed comparing mice which had SRSF2 deleted or mutated with an appropriate control.
