scRNAseq analysis for human PDX

ScRNAseq analysis was performed for human patient derived xenografts from three patients. For PDX from patient #3 (P3), stealth RNA for Negative control (Thermofisher Scientific) was transfected 24h before transplantation. PDXs were subcutaneously transplanted on 7 week old age NOG mice and grown tumor were collected 8weeks after transplantation for PDXs from Patient 1 (P1), Patient 2 (P2) or 5weeks after transplantation for P3. The collected PDXs were subjected to collagenase (WAKO) and alive cells were collected. Single alive cells were loaded into 10X Chromium Single Cell Capture Chip, followed by single-cell capture,reverse transcription and library preparation according to manufacturers' protocol. Prepared library was sequenced on the Ilumina Hiseq platform (P1, P2) or Nova Seq platform (P3), according to manufacturers' protocol.
