RNA-seq and ATAC-seq to study dendritic cell heterogeneity

The cDC2 subset is heterogeneous. We found the expression of T-bet delineates two distinct cDC2 subsets. Transcriptional profiling and analysis of chromatin accessibility in newly defined DC subsets identifies phenotypic specialisation and transcriptional regulators. ATAC-seq in cDC1s, T-bet+ cDC2 and T-bet- cDC2s (duplicates). RNA-seq in cDC1s, T-bet+ cDC2 and T-bet- cDC2s (triplicates or duplicates).

2型常规树突状细胞(conventional dendritic cell 2, cDC2)亚群具有异质性。研究发现,T-bet的表达可将cDC2亚群划分为两个特征迥异的亚群。通过对新定义的树突状细胞(dendritic cell, DC)亚群开展转录谱分析与染色质可及性分析,可鉴定其表型特化特征与转录调控因子。针对1型常规树突状细胞(conventional dendritic cell 1, cDC1)、T-bet阳性cDC2及T-bet阴性cDC2开展转座酶可及性测序(Assay for Transposase-Accessible Chromatin using sequencing, ATAC-seq),实验设置两次生物学重复。针对上述三类树突状细胞亚群开展RNA测序(RNA sequencing, RNA-seq),实验设置三次或两次生物学重复。
