To check the effectes of ST8Sia6 overexpression we performed total RNS sequesnning in porcine kidney endothelial cells to understand the biological functions of DEGs associated with ST8Sia6 overexpression in porcine kidney endothelial cells upon co-culture with human PBMCs, transcriptomic analysis was performed on four independent experimental conditions, including control GFP-expressing cells and ST8Sia6-overexpressing cells. Porcine kidney endothelial cells overexpressing ST8Sia6 and control-GFP cells were collected from four individual experiment after 24 hours transfection for total RNA extraction. Similarly, cells overexpressing ST8Sia6 and control-GFP were collected from co-culturing with four individuals PBMCs as previously described. Cells from different experimental conditions were collected keeping in ice, maintaining temperature at 4°C, washed with cold PBS, and stored as cell pellets at -80°C for total-RNA isolation.
