To understand the mechanism underlying the transcriptional regulation by TWIST1-dimers, we analysed dimer-expressing embryoid bodies by RNA-seq. We constructed dimers of TWIST1 and selected E-proteins partners: TWIST1-TWIST1 (TT), TWIST1-TCF3 (T3), TWIST1-TCF4 (T4) and TWIST1-TCF12 (T12), and also TWIST1 monomer (T0) for comparison. The coding sequences were stably integrated into the genome of A2LoxCre ESCs, downstream of a doxycycline inducible promotor (Iacovino et al., 2014). ESC lines were differentiated for three days subjected for transcriptome analysis. Protein expression were induced 16 hours before sample collection.
