Yeast RNAi libraries, YeaLKD-lib and YeaHKD-lib, constructed based on plasmid with different DNA insert, was subject to microfluidic droplet screening to enrich variant cells with high amylase production. The enriched libraries, post_sorted_YeaLKD-lib and post_sorted_YeaHKD-lib as well as YeaLKD-lib and YeaHKD-lib were then used for plasmid extraction and amplicon sequencing for the inserts respectively.
