Specialized Pro-resolving Mediators Regulate Treg Differentiation

We aim to demonstrate a central role for omega-3 derived autacoids, termed specialized pro-resolving mediators (SPM), in the differentiation and maturation of Tregs. Overall design: Naïve CD4+ T lymphocytes were isolated by immunomagnetic negative selection from the spleens of 4 WT and 4 Alox15-/- mices (KO). Tregs were differentiated from naïve CD4+ T lymphocytes for 2 days in the presence of IL-2, TGF-ß and CD3/CD28 activation beads. Using RNA sequencing we assessed the differential gene expression between KO and WT mices.
