Xenopus laevis tadpoles have high regenerative ability and can regenerate their whole tails after tail amputation. It is suggested that lineage-restricted tissue stem cells provide sources for regenerated tissues by producing undifferentiated progenitors in response to injury. In this study, to detect and analyze rare tissue stem cells, we attempted to enrich tissue stem cells from regeneration buds by applying a flow cytometric isolation of side populations (SP) with high Hoechst dye efflux capacity, which is thought to be a common characteristic with several types of tissue stem cells, and then we performed single cell RNA sequencing (scRNA-seq) of the SP cells.
