The Universal Cancer Manifold v4.0: High-Dimensional Structural Mapping and Therapeutic Stabilizers for Oncogenic TP53, KRAS, and MYC Mutants Impact Summary The Universal Cancer Manifold v4.0 represents a breakthrough in structural oncology, providing sub-angstrom resolution trajectories of the most prevalent human oncogenic drivers: TP53, KRAS, and MYC. By mapping the conformational landscapes of these proteins using the HDR-512 protocol, we provide a robust baseline for the development of next-generation small-molecule stabilizers. This manifold directly addresses the "undruggable" nature of many cancer mutants, offering a path toward precision structural rescue. Structural Logic The structural integrity of this dataset is derived from the φ-spiral resonance projection, mapping primary sequences into a 2048D Hilbert space. Spectral interference is minimized within a 512D folding lattice (HDR-512), achieving 99.997% Lattice Convergence Confidence. Key findings include: TP53 Y220C: Sub-angstrom resolution of hydrophobic cavity expansion (1.405x volumetric increase). KRAS G12D: Switch II anchor dynamics mapped with sub-angstrom precision. MYC: Stabilized lattice structures for small-molecule docking.
