From RNAseq analysis of 53 esLUAD, we identified a gene signature distinguishing invasive and indolent tumors. Regulatory network analysis identified aurora kinases as master regulators of the gene signature. So we applied an aurora kinase pan inhibitor, AMG900, using A549 and H1792 cells to confirm whether the expression of pro-invasive genes were impacted by aurora kinase activity suppression. Overall design: For RNA sequencing of A549 and H1792 cells treated with DMSO or 0.1 uM AMG900 for 48 hours. Total RNA was purified from cells using RNeasy kit (Qiagen) for sequencing performed on NextSeq 500 (Illumina).
