Analysis of transcriptional changes in cell lines (in vitro) and tumors (in vivo) after treatment with interferon a gene therapy.

Three bladder cancer cell lines were treated with recombinant IFNa, LV-Ctrl or LV-IFNa and compared with control (Ctrl) samples by RNAseq. Also, vehicle treated, LV-Ctrl and LV-IFNa treated murine MB49 bladder cancer tumor model was compared by RNAseq Overall design: Murine cell lines BBN975, MB49 and UPPL1541 were treated with recombinant IFNa (100U/ml), LV- Ctrl (MOI2) and LV-IFNa (MOI2) and compared with Ctrl samples. MB49 cells were instilled into mouse bladder and after tumor cell uptake, mice were randomized and treated with vehicle, LV-Ctrl or LV-IFNa. RNA was extracted using Ambion miRVANA kit (AM1561) and DNAase treated RNA was used to generate cDNA libraries for cell lines and murine tumors.
