Figure 4g describes the circulation half life of photoactive liposomes within the embryonic zebrafish (2dpf), before and after light triggered surface charge switching (from neutral to cationic).Confocal z-stacks were captured using a 40x water-immersion objective (HCX APO L, Leica TCS SPE microscope). Images were processed and quantified using the Fiji distribution of ImageJ. For quantification of liposome circulation lifetime decay, at least three individual embryos (biological replicates) were imaged using confocal microscopy at each time point. Quantification (not blinded), based on the mean liposome fluorescence in the lumen of the dorsal aorta, was performed on 40x confocal z-stacks (optical thickness of 2-3 µm per slice) using methods and ImageJ macros described in Campbell, F. et al. "Directing Nanoparticle Biodistribution through Evasion and Exploitation of Stab2-Dependent Nanoparticle Uptake" ACS Nano 2018, 2138-2150.
