Under adverse conditions, plants adapt to their environment through morphological structures, physiological metabolism and molecular mechanisms. In GS2, genes encoding AKT1, CNGCs, KUP family and NRT family were up-regulated to enhance potassium transport and uptake and to maintain a more stable K+ concentration and a higher K+/Na+ ratio. Metabolomics nuclear associomics analysis revealed that under low potassium stress, GS2 promotes carbohydrate synthesis, accumulates sucrose and galactose metabolism. It also enhances TCA metabolism to provides more energy. GS2 may increase the unsaturated fatty acid content, remodel membrane fluidity, and provide a structural barrier for the plant body to withstand low potassium stress. GS2 can boost the antioxidant system, accelerate the ascorbate-glutathione (ASA-GSH) cycle, and facilitate the synthesis of salicylic acid and GABA, thereby establishing a defense response. This study provides a basis for the development of high quality wild soybean resources and the extraction and use of excellent low potassium stress tolerance genes.
