Identification of calcineurin-dependent, Crz1-independent gene expression changes in response to chronic CaCl2 stress

Gene expression was quantified in S. cerevisae cells over a time course of three days of CaCl2 treatment to activate calcineurin. crz1 and crz1 cnb1 mutant strains were compared to allow the identification of calcineurin-dependent gene expression changes that are independent of the downstream transcription factor Crz1. We show that mitochondrial genes are downregulated in both geneotypes over time. Additionally, genes that promote glutathione biosynthesis are upregulated in cells that express calcineurin.

本研究对酿酒酵母(S. cerevisae)细胞在为期三天的氯化钙(CaCl2)处理以激活钙调磷酸酶(calcineurin)的时间进程中进行基因表达定量。本研究设置crz1突变株与crz1 cnb1双突变株作为对照,以鉴定不依赖下游转录因子Crz1的钙调磷酸酶依赖性基因表达变化。结果表明,两种基因型的线粒体基因均随处理时间延长呈现表达下调。此外,在表达钙调磷酸酶的细胞中,促进谷胱甘肽生物合成的基因呈现表达上调。
