The fusion of an α-helix to the N-terminus of CasMINI (hpCasMINI) could indeed enhance the DNA cleavage efficiency in mammalian cells. Overall design: To investigate the potential of hpCasMINI in genomic DNA cleavage, we transfected CasMINI or hpCasMINI or SpCas9 or LbCas12a into MCF7 cells with gRNA targeting different genomic sites. The results indicated that hpCasMINI significantly improved editing efficiency at all the tested sites compared to CasMINI.When compared to SpCas9 and LbCas12a, hpCasMINI displayed higher DNA cleavage specificity although with lower activity at the tested sites.
