Gene expression during PMA-induced neutrophil NETs formation with or without knockout of neutrophils of Src kinase

Src kinase has functions in regulating reactive oxygen species production, degranulation, NETs formation, integrin activation, and migration to human infection sites in neutrophils. RNA sequencing was performed by extracting mRNA from PMA-induced mouse neutrophils with or without Src kinase knockout(Src fl/fl S100a8 Cre or Src fl/fl mice). Gene enrichment analysis by Genomes (Reactome) identified RAF/MAP kinase cascade in the top ten significantly changed pathways. Overall design: Fresh primary neutrophils were quickly isolated from neutrophil Src kinase knockout or non knockout mice(Src fl/fl S100a8 Cre or Src fl/fl ) using the mouse bone marrow neutrophil isolation kit. NETs formation was induced by adding 100 nM Phorbol 12-myristate 13-acetate (PMA) for 4 h.
