This study utilized the tn-seq method with the HERMES transposon to determine which Candida glabrata (Nakaseomyces glabratus) genes are required for echinocandin resistance and sensitivity in strain BG2 along with a pdr1 knockout and a PDR1-Gain of Function. By growing transposon insertion pools in the presence of varying doses of micafungin, caspofungin, anidulafungin, rezafungin, and ibrexafungerp, we were able to determine which transposon insertion sites are selected for or against based on the number of reads recovered compared to an untreated control.
