Transcriptome analysis on hematopoietic progenitor cell from control and blf-/-;drl 4KO zebrafish embryo by Next Generation Sequencing

Perturbations in embryonic hematopoiesis may result in illness that ranges from fetal anemia to leukemias. We found hematopoiesis in zebrafish embryo is synergistically regulated by blf and drl cluster genes. In order to determine how loss of blf and drl cluster genes changes the transcriptome landscape, hematopoietic progenitors were collected via FACS by utilizing RFP expression from blf gene trapping allele as lineage tracer and low input RNA-seq was applied. Hematopoietic progenitors from blf+/- and blf-/-;drl 4KO embryos were collected at 20 hpf via FACS by utilizing RFP expression from blf gene trapping allele as lineage tracer. The libraries were constructed with NEBNext® Single Cell/Low Input RNA Library Prep Kit for Illumina (NEB, Cat No.6420) and sequenced on Illumina Novaseq platform.
