Overall, our studies revealed that UM171 differentially enriches specific populations of hematopoietic progenitors, in HSC expansion and lymphoid culture conditions, and that it is a potent small molecule agonist for expansion of granulocytic progenitors and lymphoid progenitors from hPSCs. Overall design: hPSC derived hematopoietic progenitors generated from H1 hESCs on day 9 of differerntiation (Uenishi et all, Stem cell Reports, 2014) expanded for 16 hrs or 5 days with UM171 were subject to gene expression profiling . DMSO treated hPSC derived hematopoietic progenitors were used as the vehicle control.
