RNA-seq of SCLC cells after treatment of cisplatin and disulfiram

Small cell lung cancer H69 cells were treated with DMSO control, 2 μM Cisplastin, 0.025 μM Disulfiram, and Cisplastin combined with Disulfiram for 24 hours. Total RNA was extracted using RNA Easy Fast Tissue/cell Kit (Tiangen, Beijing, China). Enrich mRNA using VAHTS mRNA Capture Beads (Vazyme, Jiangsu, China). Prepare the library using the VAHTS Universal V8 RNA Seq Library Prep Kit (Vazyme, Jiangsu, China), and then perform paired end sequencing on the Illumina NovaSeq 6000 machine (Illumina, CA, USA) to generate 150 bp paired end reads. Trim the original reading using Trim Galore (v0.6.7) to remove low-quality readings. Then, clean reads were aligned with the GRCh37 genome using STAR (v2.7.10b) and quantified using RSEM (v1.3.1).
