Analysis by RNA-seq of the changes in the transcriptome induced by cohesin inactivation

The main objective of this study is to determine how cohesin influences transcription and compare these changes to changes previously observed in chromatin structure, which were previously determined by Mnase-seq Overall design: Samples for RNA were obtained from two different genetic backgrounds of S.cerevisiae cells (wild-type and scc1-73-W303 bar1? background). scc1-73 is a thermosensitive allele of the kleisin subunit of the mitotic cohesin complex that was used to analyze the effects of cohesin inactivation on chromatin. 2 samples for each genetic background (2 independent experiments) were collected at the G2/M transition.
