The purpose of this study was to determine the effect of EZH2 inhibitor (EZH2i) PF-06821497 on CD45+ cells isolated from MC38 tumors at day 17 post implantation. Mice were either treated for 7 days prior to endpoint or 16 days prior to endpoint with the EZH2i or with vehicle control. CD45+ cells were isolated and scRNA-Seq was performed on cells Overall design: Mice were implanted with MC38 tumor cells on day 0 and began treatment with either vehicle control or EZH2i PF-0682497 at 100 mg/kg on day 01 for 16 day treatment group or on day 10 for 7 day treatment group. All mice were euthanized, tumors removed, digested, and CD45+ cells were enriched. Libraries were generated using Single Cell 3' GEM, Library and Gel Bead Kit v3 (10X Genomics, PN-1000075), Chromium Single Cell B Chip (PN-100073), and Chromium i7 Multiplex Kit (PN-120262). Sequencing was performed by Novogene.
