RBP-J was initially identified as a key regulator in the canonical Notch signaling pathway. More recent studies have unveiled its important role as a significant player in macrophage polarization, TLR and TNF signaling pathways. Since TNF activates RBP-J, we investigated the transcriptomic changes in murine bone marrow macrophages treated without or with TNF. Overall design: Mouse bone marrow derived macrophages from wild type control (WT) and Rbpj conditional knock out (KO) mice, in which Rbpj is specifically deleted in myeloid lineage macrophages by crossing Rbpj flox/flox mice with LysMcre mice (Rbpj f/f;LysMCre; hereafter referred to as RbpjKO), were treated without or with TNF (40ng/ml) for 2 days. The total RNA samples were obtained from WT and RbpjKO BMMs at 0 and d2 after TNF treatment. Two independent experiments were performed.
