In breast cancer metastasis soluble factors secreted by breast cancer cells trigger a cascade of events that stimulate osteoclast differentiation in the bone microenvironment. macroH2A is a unique histone variant with a C-terminal nonhistone domain and play a crucial role in modulating chromatin organization and gene transcription. To study the effect of macroH2A1.2, an isoform of macroH2A, on breast cancer-derived osteoclastogenesis, we performed gene expression array studies in wild type and mH2A1.2 knockout MDA-MB-468 cells . Total RNA was isolated from MDA-MB-468 cells in which mH2A1.2 gene have been depleted along with the wild type cells to study the differenetial regulation of various genes in each case.
