Two assays targeting 13 or 5 epigenetic age markers for chronological age estimation from semen were developed and optimized for forensic purposes. The assays are based on quantitative DNA methylation analysis through bisulfite conversion, multiplex PCR and massively parallel sequencing using the MiSeq FGx platform. An inter-laboratory validation study was performed, using DNA methylation control and semen samples to evaluate the assay’s reproducibility and sensitivity.
