Distinct functions of tissue-resident and circulating memory Th2 cells in allergic airway disease

We report RNA-seq data from distinct memory CD4+ T cell subsets in a murine model of allergic asthma. Overall design: We sensitized and challenged C57BL/6 mice with intranasal house dust mite extract to generate memory CD4+ T helper type 2 (Th2) cells. We used fluorescence-activated cell sorting to isolate memory Th2 cells, based on the expression of the canonical Th2 cell surface marker ST2, and control memory Th cells (ST2-). We sorted ST2+ and ST2- Th cells from the lung parenchyma and secondary lymphoid organs (SLO) followed by RNA isolation, cDNA synthesis, and sequencing to generate approximately 30 million of 50 bp reads per sample. We performed differential gene expression analysis based on the cuttoff of 2-fold change in expression and a false discovery rate below 0.05.
