This experiment was performed to examine transcriptional differences between differentiated (Tim3+) and stem-like (CXCR5+) WT vs. Tgfbr2 KO antigen-specific CD8 cells at day 30 after LCMV clone 13 infection. Overall design: Naïve CD8 T cells were sorted from Tgfbr2 flox/flox (WT) and dLck-Cre+ Tgfbr2 flox/flox (Tgfbr2 KO) P14 mice, and equal numbers (2000 each) of naïve WT and Tgfbr2 KO P14 cells were co-transferred into naïve C57BL/6J mice followed by LCMV clone 13 infection with transient CD4 T cell depletion. At day 30 post infection, PD-1+ Tim3+ CXCR5- differentiated subset and PD-1+ CXCR5+ Tim3- stem-like subset were sorted from WT and KO P14 cells harvested from the spleen for RNA-Seq.
