The study introduces new RiPP compounds with cytostatic (anti-tumour) properties isolated from the soil cyanobacterium Nostoc sp. CALU 546 and the respective biosynthetic gene cluster in its whole genome sequence. The compounds feature a previously unreported combination LAP structure and sactionine internal thioether bonds as well as modified proline residue. They induce S-phase cell cycle arrest followed by mitochondria-dependent apoptosis, further strengthening their therapeutic potential. The acquired data also indicates that the main compound does not primarily induce and DNA-damage or tubulin dynamics inhibition typical for many nature-derived anti-cancer drugs. Due to its novel structure and mode of action it represents an excellent candidated for preclinical evaluation.
