Functions of SMARCB1, one of the core subunits of BAF complexes, is not fully characterized in embryonic stem (ES) cells due to inability to establish ES cells from Smarcb1-deficient embryos. We conducted RNA-seq and ATAC-seq studies in combination with the CRISPR/Cas9-mediated genetic depletion to solve the problem. Consequently, we found that SMARCB1 plays a role in maintenance of pluripotency and differentiation fidelity of ES cells, which is potentially through regulation of chromatin landscapes.
