To explore the underlying mechanisms of SIS3 attenuating TGF-Ã-induced immunosuppression on NK cells, RNA-sequencing was performed on primary BM-NK cells under TGF-à stimulation with or without SIS3 pre-treatment. Overall design: BM-NK cells pre-incubated with 10µM SIS3 for 3 hours were stimulated with 5ng/ml TGF-Ã1 for another 3 hours. Then cells were harvested as Ctrl group (no stimulation nor SIS3 pre-treatment), T group (with TGF-à simulation but no SIS3 pre-treatment) and SIS group (with TGF-à simulation and SIS3 pre-treatment) and subjected for RNA sequencing.
