To identify the AvrSr35 effector we have mutagenized the American Pgt isolate 99KS76A-1 collected in 1999 in Kansas and classified as race RKQQC. Both RKQQC and Ug99 races of Pgt are avirulent on the Sr35 gene. Before sequencing, the Pgt culture was purified by single-pustule isolation and pathotyped according to North American nomenclature system. Genomic DNA from the wild-type Pgt was sequenced using three different sequencing platforms: Illumina Miseq, Roche 454, and PacBio. The raw DNA sequence data is deposited to the NCBI database under BioProject PRJNA347320 with sample accession numbers SAMN05859640 - SAMN05859643.
