Identification of prognostic-related key genes and their impact on immune response based on high-throughput RNA sequencing

Wilms tumor is the most common type of renal malignancy in children. Accumulating evidence introduces that dysregulation of genes contributes to tumorigenesis. To explore potential therapeutic targets for WT, a more detailed and comprehensive investigation of gene expression profiles is warranted. We used high-throughput RNA sequencing to analyze the mRNA expression of the WT tissues and adjacent normal tissues. Overall design: we used 8 paired WT samples and high-throughput RNA sequencing to identify differentially expressed mRNAs. The PPI network of differential genes was constructed based on the STRING database, and top 10 HUB genes were further identified. Then, a three-gene prognostic signature for WT patients was constructed by LASSO regression analysis. In addition, we analyzed the mutational characteristics of key HUB genes and predicted the effect of immunotherapy.
