This study reveals for the first time that Jaceosidin, a natural flavonoid, ameliorates sepsis-induced myocardial injury by modulating histone lactylation modification. Using a TNF-alpha induced septic mouse model, we demonstrated that Jaceosidin treatment markedly attenuated cardiac tissue damage, as evidenced by reduced serum levels of myocardial injury biomarkers. Mechanistically, Jaceosidin targeted key enzymes associated with lactate metabolism, suppressing their enzymatic activity and subsequently decreasing aberrant histone lactylation in cardiomyocytes. This inhibition rebalanced epigenetic regulation and restored mitochondrial function. Furthermore, Jaceosidin intervention mitigated sepsis-driven metabolic reprogramming by disrupting the crosstalk between lactate accumulation and histone modification. Our findings highlight the dual regulatory role of Jaceosidin in metabolic-epigenetic networks, proposing a novel therapeutic approach for sepsis-related cardiac dysfunction by targeting the lactate-histone lactylation axis.
